All you have too do is shoot a 60 second mini documentary on any subject that grabs your interest. I am the judge for the competition and I can tell you what I am looking for; something that engages me (and in 60 seconds you can tell a lot), something that is more than just a montage, I need a story as this is a mini documentary after all!
You will also need to do more than just make a great short, you will need to do some good marketing and get people to see your film as the number of likes is key to getting onto shortlist. Marketing after all is key to being a successful filmmaker. I understand your concern that people with lots of followers have an advantage there, that is true, but you will be surprised at how few actually enter these competitions. This is the way we did the FS5 competition in 2015 and there weren’t any YTers with lots of followers in the short list, in fact the winner has 27 subscribers!
